New Sequencing Platform Delivers Same-Day Whole-Genome Results

Roche (Basel, Switzerland) has introduced AXELIOS 1, a next‑generation single‑molecule sequencing platform powered by proprietary sequencing by expansion (SBX) technology. More...

The system is positioned to combine accuracy, speed, flexibility, scalability, and cost efficiency for laboratories of varied sizes. Its run setup supports both small batches and high‑throughput projects on a single instrument, with whole‑genome results available the same day in research workflows.

AXELIOS 1 employs SBX chemistry to encode DNA or RNA into measurable surrogate polymers called “Xpandomers” that are roughly 50 times longer than the original molecules. These high signal‑to‑noise reporters are read by a reusable complementary metal‑oxide‑semiconductor sensor containing millions of nanopores, enabling near real‑time base calling and analysis. Under suitable sample and library preparation conditions, workflows can accommodate reads of up to approximately 1,500 base pairs and process from sample to Variant Calling Format with ultra‑fast turnaround.

The platform is designated for Research Use Only and is not intended for use in diagnostic procedures. Its real-world capabilities have been demonstrated through launch collaborations with institutions including Hartwig Medical Foundation and Broad Clinical Labs, where studies have reported impressive accuracy and throughput, record-breaking sequencing speed, and improved cost efficiency. Early-access users have further validated the platform through proof-of-principle applications spanning oncology, genetics, infectious diseases, and other research areas.

To support implementation, Roche will offer library preparation kits and a free, open‑source bioinformatics suite (XOOS) tailored to SBX. The platform is designed for compatibility with a broad set of life‑science tools, including a partnership with 10x Genomics for single‑cell and spatial application kits and support for SBX via Google DeepVariant, facilitating flexible analysis pipelines in research environments.
