По последним оценкам, в США с диабетом живут 1,3 миллиона взрослых. Хотя причины диабета 1 типа еще не известны, существует много факторов риска для развития этого метаболического состояния. Генетические факторы риска развития T1D были идентифицированы в более чем 50 различных генетических локусах, но самой большой генетической детерминантой T1D остается область человеческого лейкоцитарного антигена (human leukocyte antigen, HLA).

Группа ученых, в том числе из Университета Линчёпинга (Linköping, Sweden), изучила данные, доступные из исследования All Babies in Southeast Sweden (ABIS). Исследование ABIS было проведено с целью выяснения, почему у детей развиваются заболевания, связанные в первую очередь с иммунной системой. Исследование ABIS содержит данные из анкет и биологических образцов более чем 17 000 детей, родившихся в 1997-1999 годах. В рамках исследования ученые собрали биологические образцы детей при рождении, в 1 год, 2–3 года и 5–6 лет. Образцы включали кровь, мочу, стул и волосы.

Чтобы провести сравнение различных уровней риска HLA на предмет развития аутоиммунного T1D, субъекты с доступными данными о генотипе HLA были отнесены к одной из четырех категорий риска на основе их генотипа HLA. ДНК из каждого образца экстрагировали из ~200 мг стула с использованием набора для экстракции  E.Z.N.A. ДНК, использовавшаяся для последующей полимеразной цепной реакции (ПЦР), была количественно определена, а ее чистота оценивалась с использованием спектрофотометра NanoDrop. Ампликоны из каждого образца были объединены и секвенированы на платформе MiSeq.

Ученые сообщили, что как микробиом ядра, так и бета-разнообразие различаются в зависимости от группы риска HLA и генотипа. Кроме того, защитные гаплотипы HLA связаны с видами бактерий Intestinibacter и Romboutsia. Было обнаружено, что у детей с высоким генетическим риском оказался другой состав кишечной микробиоты и другие кишечные микробиомы, чем у детей с низким риском. Авторы пришли к выводу, что информация, полученная от группы этого типа, способствует нашему пониманию того, как генетический риск HLA управляет изменениями в кишечном микробиоме и как генетика может заложить основу для запускающих механизмов окружающей среды, которые в конечном итоге приводят к аутоиммунному заболеваниюT1D.
