 前列腺特异抗原(PSA)是前列腺细胞产生的一种蛋白质,通过两种方式在体内循环:与其它蛋白质结合或自主行动。独自行动的PSA称为游离PSA。游离PSA检验测量未结合PSA的百分比;PSA检验测量游离PSA与结合PSA的总和。

前列腺癌会使PSA水平升高,但其它病症也会,包括前列腺肥大、前列腺炎和衰老。实际上,此前的研究显示PSA水平升高的男性约75%没有前列腺癌。以前,为了确定哪些男人有癌症而哪些没有,医生要做活组织检查。

位于无锡的江苏大学核医学研究所的科学家开发了一种新颖、灵敏的快速方法,通过联用时间分辨荧光免疫测定(TRFIA)与免疫磁性分离两种技术,可同时确定游离前列腺特异抗原(fPSA)与总前列腺特异抗原(tPSA)。新方法用磁性颗粒作为固定基质和分离手段,用发光的铕(Eu)和钐(Sm)螯合物作为探针。用tPSA和fPSA的一步式夹心TRFIA免疫测定作为血清中的分析物模型,评估了所提出的方法。

这种一步式方法不到15分钟就检测出fPSA和tPSA,fPSA的检测限是0.006纳克/毫升,tPSA的检测限是0.05纳克/毫升。fPSA和tPSA的测定范围均为0.5纳克/毫升至100纳克/毫升,而原始模式获得的平均恢复率分别为95.9%和95.3%。现在的TRFIA新方法可重复性更强,恢复率更高,灵敏度和特异性更高,事实证明它能得到广泛接受。

该研究的论文发表于2017年1月19日的《临床化验分析》(Journal of Clinical Laboratory Analysis)杂志。作者总结说,同时确定方法快速而灵敏,可置于重要位置,筛查大量样本,可通用于临床确定人血清中的fPSA和tPSA。
