По имеющимся сведениям, иммунофенотипические характеристики слизеобразующих опухолей демонстрируют значительную степень перекрывания между первичными участками локализации опухоли. Характеристики неспецифического окрашивания низкомолекулярных цитокератинов (CK7, CK20) были хорошо документированы в слизеобразующих опухолях, включая опухоли яичников, колоректального и легочного происхождения.

Патологи из Медицинской школы Университета Нью-Мексико (Альбукерке, штат Нью-Мексико, США) и их коллеги провели прямое сравнительное исследование экспрессии особого AT-богатого связывающего последовательность белка 2 (special AT-rich sequence-binding protein 2, SATB2) и гомеобокса 2 каудального типа (caudal type homeobox 2, CDX2) в разнообразных массивах первичных слизеобразующих опухолей. Иммуногистохимическое окрашивание SATB2 и CDX2 было выполнено на целых срезах из 44 слизистых колоректальных карцином и 175 неколоректальных слизистых опухолей. Была применена ядерная система подсчета, измеряющая интенсивность (0–3+) и процентное окрашивание (0 = <5%, 1 = 5–49%, 2 = ≥50%), что позволило получить результат H-score. Платформа Ventana Discovery (Ventana Medical Systems, Tucson, штат Аризона, США) использовалась для применения антител к SATB2 и CDX2.

Ученые сообщили, что SATB2 продемонстрировал приемлемую точность при низких и умеренных уровнях экспрессии (H-score 1–4). С этими пороговыми значениями H-score общая точность была более 90%. В отличие от этого, точность CDX2 превосходила точность SATB2 только при H-score, равном 5 (89,0%), поскольку его специфичность ухудшалась при более низких уровнях экспрессии (<70,0% при H-score 1–4). При использовании умеренного порогового значения H-score, равного 3 или выше, были выявлены значительные различия как по чувствительности, так и по специфичности между SATB2 и CDX2, хотя эти окрашивания были почти эквивалентными, когда каждое из них интерпретировалось как положительное при соответствующем оптимальном H-score (SATB2 ≥ 3 и CDX2 = 5).

Авторы пришли к выводу, что SATB2 является более точным маркером рака колоректального происхождения на различных уровнях экспрессии, по сравнению с CDX2, при применении к слизеобразующим опухолям из множества первичных участков локализации опухоли. Тем не менее эти окрашивания почти эквивалентны, когда каждое интерпретируется на своем оптимальном уровне экспрессии. Исследование было опубликовано в сентябрьском номере журнала “Архивы патологии и лабораторной медицины” (Archives of Pathology & Laboratory Medicine) за 2019 год.
