麝猫后睾吸虫是肠虫感染特有的,因为它引起肝内胆管里的慢性炎症反应,慢性炎症反应从晚期导管周围纤维化(APF)发展成胆管癌(CCA)。

慢性麝猫后睾吸虫感染转化为CCA的过程中,氧化应激(OS)起着重要作用,还导致修饰DNA损伤(8-oxodG)被切除,排入尿中,免疫测定可检测8-oxodG的水平。

泰国坤敬大学的科学家及来自多国的合作者把149人分成三组;第1组23人的大便检查呈麝猫后睾吸虫阴性(未感染);第2组48人呈麝猫后睾吸虫阳性(OV+)但呈APF阴性;第3组78人呈麝猫后睾吸虫阳性以及APF阳性。第4组包括33人的样本,他们经组织学检查确认有麝猫后睾吸虫相关的CCA,他们的尿样得到保存。

隔夜收集1、2、3组人员的空腹静脉血样本,装进硅化试管。收集后让静脉血样本在室温下凝结30分钟,然后离心,恢复血清。用酶联免疫吸附测定(ELISA)量化血清中麝猫后睾吸虫抗体的水平。用经过确证的竞品ELISA试剂盒测量尿液中8-oxodG的浓度,该试剂盒是HT Trevigen公司(Gaithersburg, MD, USA)的产品。用分子器件公司(Sunnydale, CA, USA)的SpectraMax 340PC384微孔板酶标仪扫描ELISA板。

尿液中8-oxodG尿液浓度的升高与APF和CCA正向相关,且与剂量密切相关。尿中8-oxodG的浓度还准确预测了一个人是否有APF或CCA,从而排除感染麝猫后睾吸虫但没有上述病症的人。尿液8-oxodG是一种可靠的候选生物标记物,可预报从慢性后睾吸虫症向APF和CCA的演进,说明氧化应激对这两种晚期肝胆病都起着重要作用。
