木瓜叶提取物可以减慢癌细胞培养物的生长

癌症研究人员发现,使用一种从干木瓜叶中制备而来的水提取物对10多种不同种类肿瘤细胞培养物进行试验时,有很强的抗癌特性。

佛罗里达大学(Gainesville, FL, USA; www.ufl.edu)和东京大学(Japan; www.u-tokyo.ac.jp)的研究人员试图利用科学来研究木瓜的抗癌特性。他们的计划是,研究一种水提取的木瓜叶成份对各种肿瘤细胞株生长的作用和对人体淋巴细胞抗肿瘤作用的影响。另外,他们还尝试找出木瓜叶提取物中的功能分子量分值。在这项研究中,木瓜叶提取物对肿瘤细胞株和人体外周血单个核细胞(PBMC)的增生反应的作用以及对PBMC细胞毒素活性的作用是通过使用放射性胸腺嘧啶来进行评估的。他们还进行了流式细胞检测分析和半胱氨酸天冬酶-3/7活性测量来证实肿瘤细胞发生了细胞凋亡。PBMC产生的细胞因子是通过ELISA来衡量的。叶子提取物治疗效果的基因图谱是通过微阵列分析和实时RT-PCR进行的。

公布在2010年2月17号《民族药理学杂志》上的结果揭示出,木瓜叶提取物对宫颈癌,乳腺癌,肝,肺,胰腺肿瘤细胞株有明显生长抑制活性的作用。在PBMC中,加入木瓜叶提取物后白细胞介素-2(IL-2)和IL-4的生成减少,同时,IL-12p40, IL-12p70, IFN-γ(干扰素-γ)和TNF-α(肿瘤坏死因子-α)的生长增加,没有受到生长抑制。另外,活性PBMC的细胞毒性在加入提取物后增加。而且,微阵列分析显示,23种免疫调节基因的表达,根据基因本体论分析分类,在加入木瓜叶提取物后升高。

研究人员识别出了木瓜叶提取物的活性成份,会抑制肿瘤细胞生长和刺激抗肿瘤作用,这种成份的分子量小于1000。重要的是,木瓜提取物不会对正常细胞有任何有毒作用。

"根据我在临床情况中所看到的和所听到的,没有人使用这种提取物出现明显的毒副作用;似乎可以使用很长时间——只要这种提取物有效的话",佛罗里达大学血液学和肿瘤学教授、供稿作者Nam Dang博士说。
