用经过亲水处理的塑料板诊断疟疾

疟疾是由疟原虫引起的一种红细胞(RBC)感染,红细胞感染率对于研究和诊断疟疾至关重要,可通过显微镜检查玻片上的吉姆萨染色薄血液涂片确定红细胞感染率。

然而,血液涂片上只有一小块适合用以确定感染率的单层红细胞,这是吉姆萨显微镜检检测寄生虫感染的准确率低的主要原因之一。此外,因为吉姆萨显微镜检条件严苛且耗时,所以亟需自动测量感染率。

日本东京国家先进工业科技研究所(AIST)的科学家培养了一种恶性疟原虫,以自动计数寄生虫感染;用可透过细胞的绿荧光核酸染料对感染寄生虫的红细胞染色。用德国韦茨拉尔市徕卡微系统公司的DM1L倒置荧光显微镜获得经染色的感染寄生虫红细胞的明场图像和荧光图像。

研究人员开发了一种方法,用显微镜检查几乎铺满经过亲水处理的环烯烃共聚物(COC)板整个表面的单层吉姆萨染色细胞。因为能在经过亲水处理的板上执行多种多样的吉姆萨显微镜检,所以该方法也许能更可靠地诊断低寄生虫血症患者体内的疟疾。还能用荧光核染料对红细胞和寄生虫染色,并用软件工具自动计数,无需吉姆萨染色。结果,检查疟疾的医务人员能轻松、快速、准确地计算感染率,即使是低寄生虫血症。

该研究的论文发表于2017年8月8日的《疟疾杂志》(Malaria Journal)。作者总结说,这些方法的运行成本极低,不涉及复杂技术,而且使用亲水的COC板也许有助于更好、更准确地诊断疟疾。
