ДНК ротовых бактерий, в основном из группы viridans streptococci (малопатогенные стрептококковые бактерии), была обнаружена в аспиратах коронарного тромба пациентов, перенесших инфаркт миокарда и аневризмы сосудов головного мозга. Тромбоз мозговой артерии вызывает 87% инсультов. Большинство тромбов формируются в стенозах сонной артерии, откуда они перемещаются, чтобы блокировать мозговое кровообращение.

Ученые из Университета Тампере (University of Tampere; Тампере, Финляндия) проанализировали аспираты тромба, которые нейроинтервационные радиологи удалили у 75 пациентов с инсультом (69% мужчин, средний возраст 67 лет) в рамках неотложной терапии. Бактериальная ДНК была извлечена из образцов с использованием коммерчески доступного набора QIAamp DNA Mini Kit.

Универсальные бактериальные праймеры и зонды были определены с помощью RNaseP. Присутствие видов Streptococcus, в основном группы Streptococcus mitis, принадлежащих к viridans streptococci, а также Porphyromonas gingivalis и Aggregatibacter actinomycetemcomitans в образцах определяли с помощью количественной полимеразной цепной реакции (кПЦР) со специфическими праймерами и зондами. Анализы кПЦР проводили с использованием специфической гибридизации аллеля TaqMan с секвенатором Abi Prism 7900 HT.

Команда сообщила, что бактериальная ДНК была обнаружена в 63 (84%) аспирированных тромбах, и 12 (16%) образцов были признаны отрицательными на бактериальную ДНК. ДНК видов Streptococcus, в основном группы S. mitis, была обнаружена в 59 (79%) образцах. Среднее относительное количество ДНК видов Streptococcus оказалось более чем в 5 раз выше по сравнению с контрольными образцами крови тех же пациентов. Все тромбы были отрицательными как на P. gingivalis, так и на A. actinomycetemcomitans.

Авторы пришли к выводу, что они обнаружили ДНК видов Streptococcus, в основном группы S. mitis, принадлежащих к viridans streptococci, в большинстве аспирированных тромбов у пациентов с острым ишемическим инсультом (ОИИ). Это говорит о том, что viridans streptococci может играть роль в возникновении цереброваскулярных заболеваний.
