研究发现乳腺癌脑转移的基因标记物

乳腺癌向脑的转移取决于若干预后因子,如年纪更轻、种族和激素受体的阴性状态。

其它因素有:BRCA1种系突变,以及表皮生长因子受体2 (Her2/neu)原癌基因的表达,二者都致使脑转移率增加。

美国亚利桑那州凤凰城转化基因组学研究所的科学家在事后从其它科研机构获得了35份新鲜冷冻的乳腺癌脑转移样本以及无肿瘤的脑组织样本和10份无肿瘤的乳腺组织样本。还获得了50份1度和2度的早期乳腺癌样本。

用凯杰公司(Valencia, CA, USA)的DNeasy血液和组织试剂盒从新鲜冷冻组织中分离基因组DNA (gDNA),用美国德克萨斯州奥斯汀市Ambion公司的mirVana miRNA分离试剂盒分离总核糖核酸(RNA),包括小RNA。在安捷伦科技公司(Santa Clara, CA, USA)的2400型生物分析仪上用生物分析仪RNA 6000纳米芯片实验室试剂盒评估RNA样本的完整性。科研小组在收集到35份乳腺癌脑转移样本后进行了深度基因组测序,整合了基因拷贝数、基因表达和DNA甲基化数据集。

该研究发现的一些常见基因变异是8号染色体里的增益和损失,还有基因变异致癌的关键机制——细胞增殖和细胞周期进展,它们与极光激酶A (AURKA)、极光激酶B (AURKB)和叉头框蛋白M1 (FOXM1)的基因有关。该研究的论文发表于2014年1月29日的《公共科学图书馆》(Public Library of Science One)杂志。作者总结说,乳腺癌脑转移的基因组和表观基因组图谱深入揭示了导致这种病的身体事件,这有望形成将来治疗策略的基础。PAM50乳腺癌内部分类器认定Luminal B、Her2+/ER阴性和基底样肿瘤是脑转移人群中最有代表性的乳腺癌亚型。
