巴西圣保罗Butantan研究所的科学家用单链可变片段(scFv)方法开发了一种重组抗体。他们用一种免疫荧光测定评估了一种紧密素重组抗体(scFv-intimin)的灵敏度和特异性。他们用scFv-intimin设计了一种合成基因。紧密素是分子量为94千道尔顿的外膜蛋白,由紧密素粘附蛋白基因(eae)编码。

作为免疫荧光对照的还有抗组氨酸小鼠抗体和山羊抗小鼠免疫球蛋白G-异硫氰酸荧光素(IgG-FITC, Sigma-Aldrich; St. Louis, MO, USA)联接体。在所有分离物中,首先用PCR测定eae基因,再用荧光肌动蛋白染色分析确认eae阳性分离物。细菌悬浮液经scFv-intimin再悬浮后,取一滴滴在玻片上,用德国慕尼黑Axioskop-Zeiss公司的荧光显微镜观察。

scFv-intimin免疫测定检测典型EPEC、非典型EPEC和EHEC分离物,灵敏度为100%,在eae阴性分离物中没有检测到细菌,说明特异性为100%。该研究的论文发表于2013年10月2日的《诊断微生物学与传染病》(Diagnostic Microbiology and Infectious Disease)杂志。作者总结说,免疫荧光是一种有效而快速的方法,scFv-intimin既是诊断EPEC和EHEC所致腹泻的优良工具,又可用于病例-对照流行病调查。
