Чтобы решить эту прогностическую задачу, исследователи из Университета Джонса Хопкинса (Johns Hopkins University; Балтимор, штат Мэриленд, США) разработали микрофлюидный анализ, который количественно определил численность и пролиферативный индекс мигрирующих клеток в образцах рака молочной железы для оценки их метастатической склонности и для быстрого скрининга потенциальных антиметастатических препаратов.

Устройство содержало Y-образные микроканалы, размеры которых подобраны для имитации аспектов сложности и разнообразия областей поперечного сечения следов ткани, найденных в на различных участках тела, или вдоль них. Этот микрофлюидный анализ для количественной оценки инвазии клеток (Microfluidic Assay for quantification of Cell Invasion - MAqCI) был разработан для оценки трех ключевых характеристик метастазирования: способности раковых клеток двигаться, способности сжиматься, чтобы проникать в узкие каналы, и способности размножаться.

Устройство MAqCI, которое в настоящее время запатентовано в Соединенных Штатах Америки, использовалось совместно с компьютеризированным инвертированным микроскопом, обладающим особенностями фазового контраста и флуоресцентного изображения. Миграцию клеток контролировали в режиме реального времени с помощью фазово-контрастной микроскопии. Результаты экспериментов, проведенных с устройством MAqCI, показали, что, по сравнению с несортированной популяцией раковых клеток, высокомобильные клетки, выделенные с помощью устройства, проявляли сходный онкогенный потенциал, но заметно увеличивали склонность к метастазированию in vivo.

РНК-секвенирование высокоподвижных клеток выявило обогащение генов, связанных с подвижностью и выживанием.

"Когда в теле пациента обнаруживается припухлость, врач с помощью биопсии может определить, является ли масса доброкачественной или злокачественной, но он не может с уверенностью сказать, будет ли злокачественная опухоль высокоагрессивной и метастазирующей в другие места, — сказал старший автор доктор Константинос Константопулос (Konstantinos Konstantopoulos), профессор биоинженерии и онкологии в Университете Джонса Хопкинса. — Несмотря на то, что жидкостные биопсии или измерения циркулирующей опухолевой ДНК могут быть очень хороши при мониторинге реакции пациента на терапию после ее применения, они не обеспечивают средства, чтобы помочь врачам выбрать оптимальные препараты для предотвращения распространения".
