检测方法基于以包含具体SNP的弱结合DNA双链的置换反应进行检测。通过检测石墨烯半导体管中链置换诱导抗性(因此产生的电流)的变化检测单一错配。使用大的双螺旋DNA链(多达47个碱基)将检测出假阳性结果的可能性最小化,改善SNP检测的准确性。
“单链DNA探针不能提供此选择性——甚至含有一个错配核苷酸碱基的一条DNA链能与探针结合并产生假阳性结果,”资深作者,University of California(San Diego)的生物工程、机械工程学和材料科学教授,Ratnesh Lal博士说明。“我们期望使用更长的探针,能开发一种可靠的序列专属性SNP检测芯片。事实上,我们开发的技术有高灵敏度和专属性。”
