Adaptive Biotechnologies(South San Francisco,CA,USA)的科研人员对5名T细胞低至百万分之一情况的患者进行了多重30种不同抗原检测,并鉴别出427个抗原特异性克隆型。克隆型鉴别结果得到多种途径的验证,包括重复性、与已发表克隆型的一致性和与酶联免疫斑点技术(ELISPOT)的高度相关性。
使用下列两种方法之一鉴别抗原特异性T细胞:与多肽混合物孵化过夜后进行dextramer结合或CD137上调。Dextramer特异性T细胞被孵化外周血单核细胞(PBMC)识别,结合了8个dextramer。新检测方法被称为MIRA,用于T细胞受体抗原特异性多重鉴别。
